Spencer is the only child of comedian/actor Kelsey Grammer and dance instructor Doreen Alderman. She is Grammer's oldest child, and has three younger siblings; two half-sisters and a half-brother.

Spencer got into acting at a young age, appearing on her father's show Cheers when she was just a toddler. In more recent years, Spencer joined the cast of the long-running daytime soap opera As The World Turns as medical intern Lucy Montgomery. She played this role for just over six months before exiting on December 14th, 2006 to pursue the role of Casey Cartwright in the hit television show Greek that appears on ABC Family. On the show, Spencer plays a sorority member who doesn't want her brother to become part of her college world.

Also in her career, Spencer has made numerous television appearances on shows such as Law & Order: SVU and Third Watch, and has appeared in various movies such as 2006's The Path of Most Resistance and most recently, Descent starring Rosario Dawson.
